Metabolism of C-16-oxygenated steroids by human placenta: the formation of estriol.
نویسنده
چکیده
Two distinct pathways for the formation of estriol in vitro have recently been described: C-16 hydroxylation of estradiol-17-P by human fetal liver and rat liver (1, 2), and aromatization of A5-androstene-3/?, 16a, 17&triol by human placenta (3). Although the formation in viva of estriol from estradioll and estrone is well known (4), the studies mentioned above present the first convincing data for the direct production of estriol in vitro. The preliminary observation that estriol could be produced by aromatization of a C-16-hydroxylated androgen without involving the classic conversion from estradiol (3), prompted this more extensive study of the metabolism of C-16oxygenated neutral and phenolic steroids by human placenta. The formation of estriol from C-16-hydroxylated androgens has been confirmed and a tentative intermediary role for 16ol-hydroxyestrone can now be made.

Metabolism of synthetic steroids by the human placenta.
Pregnant women with asthma are frequently exposed to synthetic glucocorticoids and glucocorticoids are known to reduce fetal growth. The fetus is normally protected from the harmful effects of maternally derived glucocorticoids by the placental enzyme 11beta-hydroxysteroid dehydrogenase type 2 (11beta-HSD2). Whether 11beta-HSD2 inactivates the synthetic glucocorticoids used for asthma treatment...
متن کامل[The metabolism and conjugation of estrogens in the human fetus].
Although several studies have been reported on the synthesis of steroids in the fetus, the metabolism and conjugation of estrogens in the fetus is not completely known. In this study serum levels of estetrol (E4), estetrol glucosiduronate (E4-G), estriol (E3) and estriol-16-glucosiduronate (E3-16-G) were measured by R.I.A. in maternal peripheral vein blood (MPV), umbilical artery blood (UA) and...
متن کاملEnzymes involved in the formation and transformation of steroid hormones in the fetal and placental compartments.
Human fetal and placental compartments have all the enzymatic systems necessary to produce steroid hormones. However, their activities are different and complementary: the fetus is very active in converting acetate into cholesterol, in transforming pregnanes to androstanes, various hydroxylases, sulfotransferases, whilst all these transformations are absent or very limited in the placenta. This...
متن کاملThe effects of the phosphorothioate insecticide fenitrothion on mammalian cytochrome P450-dependent metabolism of estradiol.
Phosphorothioate insecticides, such as fenitrothion, are suicide substrates of cytochromes P450 (P450). These compounds undergo oxidative desulfuration by P450 resulting in the release and subsequent binding of atomic sulfur to the enzyme. Consequently, the P450-dependent metabolism of certain endogenous substrates could be inhibited by exposure to these insecticides. Formation of 2-hydroxyestr...
متن کاملA Brief Account of the Discovery of the Fetal/Placental Unit for Estrogen Production in Equine and Human Pregnancies: Relation to Human Medicine
The role of steroids in human medicine is well recognized, but the major contributions made by the large domestic animals as a source of material in the discovery, isolation, and determination of the structure of the steroid hormones is less well appreciated. After a brief reminder of the early efforts to obtain a reliable source of steroids for clinical use, the narrative here is to outline on...
